RNF114 also known as zinc finger protein 313 is a RING-type E3 ubiquitin ligase with a mass of approximately 51 kDa. It plays a mechanical role in tagging proteins for ubiquitination marking them for degradation by the proteasome. RNF114 is expressed mainly in the skin but can also be found in immune cells such as T-cells and macrophages. Its E3 ubiquitin ligase activity is important for regulating protein stability and cellular processes.
Biological function summary

RNF114 has a significant role in regulating immune response and cell proliferation. It participates in pathways that control inflammation by modulating the activity of NF-kB an important transcription factor in immune response. RNF114 also influences cell cycle regulation through its interactions with other proteins. While it does not form a strict complex it interacts closely with substrates it ubiquitinates to affect signaling pathways critical for cellular responses.

Pathways

Multiple cellular processes engage RNF114. It plays a part in the NF-kB signaling pathway and the Wnt signaling pathway. In the NF-kB signaling pathway RNF114 helps control the stability of specific proteins maintaining a balance in immune responses. In the Wnt pathway it has been indicated to interact with proteins like β-catenin influencing gene expression that governs cell growth and differentiation both important in development and immune function.

A few studies connect RNF114 with autoimmune diseases and cancer. For example its dysregulation has associations with psoriasis an immune-mediated disease leading to excessive skin cell proliferation. Dysregulation of RNF114 has also been noted in certain cancer types possibly through interactions with proteins such as tumor suppressors affecting pathways regulating cell growth and death. Understanding these interactions can provide insights into therapeutic targets for these conditions.

E3 ubiquitin-protein ligase that promotes the ubiquitination of various substrates (PubMed : 23645206, PubMed : 25165885). In turn, participates in the regulation of many biological processes including cell cycle, apoptosis, osteoclastogenesis as well as innate or adaptive immunity (PubMed : 25165885, PubMed : 28708287). Acts as a negative regulator of NF-kappa-B-dependent transcription by promoting the ubiquitination and stabilization of the NF-kappa-B inhibitor TNFAIP3 (PubMed : 25165885). May promote the ubiquitination of TRAF6 as well (PubMed : 28708287). Acts also as a negative regulator of T-cell activation (PubMed : 25165885). Inhibits cellular dsRNA responses and interferon production by targeting MAVS component for proteasomal degradation (PubMed : 25165885). Ubiquitinates the CDK inhibitor CDKN1A leading to its degradationand probably also CDKN1B and CDKN1C (PubMed : 23645206). This activity stimulates cell cycle G1-to-S phase transition and suppresses cellular senescence. May play a role in spermatogenesis.
