Supplementary information

RNF138 also known as HSD-4 or RING finger protein 138 acts as an E3 ubiquitin-protein ligase. It influences protein degradation by attaching ubiquitin to target proteins signaling them for proteasomal degradation. This protein typically has a molecular mass of approximately 28 kDa. RNF138 is broadly expressed in various tissues but often shows higher expression in neuronal tissues and skin. The presence of a RING finger domain enables RNF138 to interact with substrates and E2 conjugating enzymes directly.
Biological function summary

The function of RNF138 plays a significant role in DNA damage repair and neuronal development. It often operates within a larger protein complex collaborating with other RING finger proteins and enzymes involved in ubiquitination. By facilitating the removal of damaged or excess proteins RNF138 supports cellular homeostasis. Its specific activity in neurons suggests a vital involvement in neural differentiation and axonogenesis processes essential for healthy nervous system development.

Pathways

Multiple cellular networks integrate RNF138 function including the ubiquitin-proteasome pathway and the DNA damage response pathway. It interacts with key proteins like BRCA1 and RAD51 modulating their stability and function. The involvement of RNF138 in these pathways highlights its role in maintaining genetic stability and regulating cellular responses to stress. It ensures proper cellular function by influencing protein turnover rates important for cell cycle and repair mechanisms.

Researchers associate RNF138 dysfunction with neurological disorders and certain types of cancers. Aberrant expression or mutation in RNF138 can lead to improper ubiquitination processes contributing to tumorigenesis linked with proteins like p53 and MDM2. In neurodegenerative diseases its altered activity might affect neuron survival due to disrupted protein homeostasis. Understanding RNF138 interactions provides potential therapeutic targets for mitigating these disorders' impact.

Target data

E3 ubiquitin-protein ligase involved in DNA damage response by promoting DNA resection and homologous recombination (PubMed : 26502055, PubMed : 26502057). Recruited to sites of double-strand breaks following DNA damage and specifically promotes double-strand break repair via homologous recombination (PubMed : 26502055, PubMed : 26502057). Two different, non-exclusive, mechanisms have been proposed. According to a report, regulates the choice of double-strand break repair by favoring homologous recombination over non-homologous end joining (NHEJ) : acts by mediating ubiquitination of XRCC5/Ku80, leading to remove the Ku complex from DNA breaks, thereby promoting homologous recombination (PubMed : 26502055). According to another report, cooperates with UBE2Ds E2 ubiquitin ligases (UBE2D1, UBE2D2, UBE2D3 or UBE2D4) to promote homologous recombination by mediating ubiquitination of RBBP8/CtIP (PubMed : 26502057). Together with NLK, involved in the ubiquitination and degradation of TCF/LEF (PubMed : 16714285). Also exhibits auto-ubiquitination activity in combination with UBE2K (PubMed : 16714285). May act as a negative regulator in the Wnt/beta-catenin-mediated signaling pathway (PubMed : 16714285).
