Supplementary information

RNF183 also known as RING finger protein 183 is an E3 ubiquitin ligase. It is approximately 28 kDa in mass and belongs to the RING finger protein family characterized by a specific motif for binding zinc ions. RNF183 directs the transfer of ubiquitin from E2 ubiquitin-conjugating enzymes to specific substrates marking these proteins for proteasomal degradation or altering their cellular functions. This protein is expressed mainly in the kidney and small intestine with some presence in other tissues.
Biological function summary

The RNF183 protein regulates protein degradation cellular signaling and stability. It often acts independently and not as part of a larger protein complex influencing various cellular processes. An important function includes regulating pathways associated with immune responses and inflammatory signaling essential for maintaining cell homeostasis and immune system balance. Through its activity RNF183 modulates specific proteins involved in cellular signaling cascades.

Pathways

RNF183 mainly participates in ubiquitin-proteasome and inflammatory response pathways. It interacts with other proteins such as TNF receptor-associated factors which are important for relaying information to downstream signaling molecules. In the context of the NF-kB signaling pathway RNF183 influences this important immune pathway by dictating the stability and activity of various signaling proteins affecting immune response and inflammation regulation.

Studies have linked altered RNF183 expression to digestive system disorders and certain cancers such as gastric cancer. Changes in RNF183 levels can disrupt normal cellular signaling contributing to tumorigenesis and disease progression. The interaction with TRAF proteins and other components within the affected pathways underlines its role in disease mechanisms providing potential therapeutic targets for research and drug development.

Target data

Acts as an E3 ubiquitin ligase catalyzing the covalent attachment of ubiquitin moieties onto substrate proteins (PubMed : 29507230). Triggers apoptosis in response to prolonged ER stress by mediating the polyubiquitination and subsequent proteasomal degradation of BCL2L1 (PubMed : 29507230). May collaborate with FATE1 to restrain BIK protein levels thus regulating apoptotic signaling (PubMed : 26567849).
