Supplementary information

RNF185 also known as RING finger protein 185 is an E3 ubiquitin-protein ligase that plays an important role in the ubiquitination process. It works with RNF5/NG2 another protein and both belong to the RING finger protein family which is characterized by a specific domain that facilitates protein-protein interactions. This protein has a mass of approximately 35 kDa. RNF185 is expressed across various tissues with higher levels often noted in the heart and skeletal muscle indicating a role in cell function within these tissues.
Biological function summary

RNF185 regulates protein homeostasis by marking target proteins for degradation via ubiquitination. It does not act alone; often it forms part of a larger complex that includes components like E2 ubiquitin-conjugating enzymes. This activity ensures that cells maintain the proper levels of proteins helping remove misfolded or damaged proteins. The complex activity of RNF185 is essential for cellular quality control mechanisms maintaining balance within the protein environment of the cell.

Pathways

Involvement of RNF185 connects it to important cellular processes including the endoplasmic reticulum-associated degradation (ERAD) pathway and the Wnt signaling pathway. In ERAD RNF185 collaborates with other proteins such as RNF5 to facilitate degradation of misfolded proteins in the endoplasmic reticulum. Through the Wnt pathway control of protein levels by RNF185 influences cellular proliferation and differentiation processes that are essential for proper tissue development and homeostasis.

RNF185 shows significant links to cardiovascular diseases and some forms of cancer. Dysregulation of RNF185 and its interaction with proteins like RNF5 can lead to abnormal protein accumulation or degradation contributing to pathogenesis in these conditions. In cardiovascular diseases improper protein turnover affecting heart and muscle tissues can lead to cardiac dysfunction. In cancer altered activity of RNF185 may affect cell growth pathways promoting tumor growth and progression.

Target data

E3 ubiquitin-protein ligase that regulates selective mitochondrial autophagy by mediating 'Lys-63'-linked polyubiquitination of BNIP1 (PubMed : 21931693). Acts in the endoplasmic reticulum (ER)-associated degradation (ERAD) pathway, which targets misfolded proteins that accumulate in the endoplasmic reticulum (ER) for ubiquitination and subsequent proteasome-mediated degradation (PubMed : 27485036). Protects cells from ER stress-induced apoptosis (PubMed : 27485036). Responsible for the cotranslational ubiquitination and degradation of CFTR in the ERAD pathway (PubMed : 24019521). Also acts as a regulator of the innate antiviral response by catalyzing 'Lys-27'-linked polyubiquitination of CGAS at 'Lys-173' and 'Lys-384', thereby promoting CGAS cyclic GMP-AMP synthase activity (PubMed : 28273161). Preferentially associates with the E2 enzymes UBE2J1 and UBE2J2 (PubMed : 24019521).
