E3 ubiquitin-protein ligase that regulates selective mitochondrial autophagy by mediating 'Lys-63'-linked polyubiquitination of BNIP1 (PubMed:21931693). Acts in the endoplasmic reticulum (ER)-associated degradation (ERAD) pathway, which targets misfolded proteins that accumulate in the endoplasmic reticulum (ER) for ubiquitination and subsequent proteasome-mediated degradation (PubMed:27485036). Protects cells from ER stress-induced apoptosis (PubMed:27485036). Responsible for the cotranslational ubiquitination and degradation of CFTR in the ERAD pathway (PubMed:24019521). Also acts as a regulator of the innate antiviral response by catalyzing 'Lys-27'-linked polyubiquitination of CGAS at 'Lys-173' and 'Lys-384', thereby promoting CGAS cyclic GMP-AMP synthase activity (PubMed:28273161). Preferentially associates with the E2 enzymes UBE2J1 and UBE2J2 (PubMed:24019521).

Activity summary

The RNF185 and RNF5/NG2 proteins function as E3 ubiquitin ligases which add ubiquitin to substrate proteins and mark them for degradation by the proteasome. Together these proteins play an important role in the endoplasmic reticulum-associated degradation (ERAD) pathway. RNF185 is also known as Ring Finger Protein 185 while RNF5 is often referred to as Ring Finger Protein 5 or NG2 in some contexts. RNF185 has a mass of approximately 34 kDa and RNF5 has a mass of about 22 kDa. These proteins express primarily in tissues such as the heart liver and kidneys participating in cellular quality control and stress responses.

Biological function summary

These ubiquitin ligases mediate protein homeostasis by regulating the turnover of misfolded or damaged proteins through the ubiquitin-proteasome system. They interact with several other proteins to form a complex involved in tagging defective molecules. This complex helps maintain cellular integrity by preventing the accumulation of potentially toxic proteins. RNF185 and RNF5 also regulate cellular responses under stress conditions revealing their intervention in adaptive mechanisms of cells to maintain survival when under duress.

Pathways

RNF185 and RNF5 coordinate within the ERAD pathway and contribute to protein quality control by linking recognition of misfolded proteins to proteasomal degradation. They relate closely with proteins such as Ubiquitin-Conjugating Enzyme E2 (UBE2) and Derlin-1 integral players in the ERAD process. Their participation in this pathway is fundamental for removing defective proteins therefore upholding cellular function in stress environments. Both RNF185 and RNF5 influence pathways like the Unfolded Protein Response (UPR) further highlighting their importance in maintaining protein balance and cellular health.

Associated diseases and disorders

Defects or dysregulation in RNF185 and RNF5 expression have implications in conditions like neurodegenerative diseases and cancer. Aberrant activity of these proteins may lead to the accumulation of misfolded proteins or disruption of normal cellular processes contributing to disease pathology. For instance in the context of cancer these proteins interact with components such as the tumor suppressor protein p53 affecting cell cycle regulation and apoptosis. Their involvement in neurodegenerative disorders may be linked to erroneous removal of proteins contributing to neuronal damage and disease progression.
