Biological function summary
RNF216 functions in protein homeostasis and immune system regulation. It acts as a mediator in the disposal of misfolded proteins preventing accumulation that could lead to cellular stress. RNF216 does not work alone; it often interacts with E2 ubiquitin-conjugating enzymes to facilitate substrate targeting and ubiquitination. Furthermore RNF216 is essential for innate immune response modulation through degradation of immune stimulatory proteins.
Pathways
RNF216 takes part in the protein degradation pathway and immune signaling pathways. In the ubiquitin-proteasome system RNF216 collaborates with E2 enzymes as an important player in proteolysis. It also plays a role in regulating the NF-kB pathway by affecting the levels of proteins necessary for NF-kB activation such as TRAF3. These interactions ensure that response to stress and infections is tightly controlled which otherwise could lead to inflammatory conditions.
