Supplementary information

The molecular structure RNF216 also known as Ring Finger Protein 216 and TRIAD3A is an E3 ubiquitin-protein ligase. RNF216 weighs approximately 101 kDa and contains a RING finger domain which is important for its ligase activity. This protein labels specific target proteins with ubiquitin marking them for degradation by the proteasome. RNF216 can be found in various tissues in the body indicating a widespread expression but it is significantly present in neural tissues.
Biological function summary

RNF216 functions in protein homeostasis and immune system regulation. It acts as a mediator in the disposal of misfolded proteins preventing accumulation that could lead to cellular stress. RNF216 does not work alone; it often interacts with E2 ubiquitin-conjugating enzymes to facilitate substrate targeting and ubiquitination. Furthermore RNF216 is essential for innate immune response modulation through degradation of immune stimulatory proteins.

Pathways

RNF216 takes part in the protein degradation pathway and immune signaling pathways. In the ubiquitin-proteasome system RNF216 collaborates with E2 enzymes as an important player in proteolysis. It also plays a role in regulating the NF-kB pathway by affecting the levels of proteins necessary for NF-kB activation such as TRAF3. These interactions ensure that response to stress and infections is tightly controlled which otherwise could lead to inflammatory conditions.

RNF216 is linked to neurological disorders and immune system dysregulation. Mutations or dysregulation of RNF216 can contribute to Gordon Holmes syndrome which involves movement and endocrine problems due to neural dysfunction. RNF216 also affects immune-related conditions by interacting with proteins like TRAF6 which play a role in immune signaling. Understanding RNF216's role provides insights into potential therapeutic targets for managing such diseases.

Target data

E3 ubiquitin ligase which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, and then transfers it to substrates promoting their ubiquitination. Plays a role in the regulation of antiviral responses by promoting the degradation of TRAF3, TLR4 and TLR9. In turn, down-regulates NF-kappa-B and IRF3 activation as well as beta interferon production. Participates also in the regulation of autophagy by ubiquitinating BECN1 leading to its degradation and autophagy inhibition. Plays a role in ARC-dependent synaptic plasticity by mediating ARC ubiquitination resulting in its rapid proteasomal degradation (By similarity). Plays aso an essential role in spermatogenesis and male fertility (PubMed : 30649198). Mechanistically, regulates meiosis by promoting the degradation of PRKACB through the ubiquitin-mediated lysosome pathway (PubMed : 33724554). Modulates the gonadotropin-releasing hormone signal pathway by affecting the stability of STAU2 that is required for the microtubule-dependent transport of neuronal RNA from the cell body to the dendrite (PubMed : 37439148).
