Protein kinase which is a key regulator of actin cytoskeleton and cell polarity. Sandwich ELISA of ROCK2 Antibody Pair - BSA and Azide free ab322250 with the capture antibody Anti-ROCK2 antibody [EPR24575-76] - BSA and Azide free (Capture) ab322251 dilution at 2 ug/mL and detector antibody ab322252 dilution at 0.5 ug/mL.
Interpolated concentrations of native ROCK2 in human control wild type HAP1 (human near-haploid cell) and ROCK2 knockout HAP1 cell based on 500 ug/mL extract loads. The concentrations of ROCK2 were measured in duplicate and interpolated from the ROCK2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ROCK2 concentration was determined to be 4191.8 pg/mL in wild type HAP1 extract and undetectable in ROCK2 knockout HAP1 extract.
Interpolated concentration of native ROCK2 was measured at different sample concentrations. Undiluted samples are as follows: HAP1 cell extract (500 ug/mL), NIH/3T3 cell extract (125 ug/mL) and C6 cell extract (125 ug/mL). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in antibody diluent CP2 (Antibody Diluent CP2 ab319981). Data represented here are indicative of assay working using example of biological sample. However, we do not guarantee same sensitivity level due to biological variation.
