Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human lung carcinoma tissue sections labeling ROS1 with Purified ab189925 at 1 : 300 dilution (5.23 μg/ml). Heat mediated antigen retrieval was performed using EDTA Buffer, pH9.0. Tissue was counterstained with hematoxylin. ImmunoHistoProbe one step HRP Polymer (ready to use) secondary antibody was used at 1 : 0 dilution. PBS instead of the primary antibody was used as the negative control.

Immunohistochemical analysis of paraffin-embedded Human lung carcinoma tissue labeling ROS1 with unpurified ab189925 at 1/250 dilution followed by prediluted HRP Polymer for Rabbit IgG. Counter stained with hematoxylin. (Inset : negative control).

Perform heat mediated antigen retrieval with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.

Blocking buffer and concentration : 5% NFDM/TBST Diluting buffer and concentration : 5% NFDM /TBST We advise using U-118MG (FIG-ROS1 fusion) or HCC78 (SLC34A2-ROS1 fusion) as a positive cell line by WB. We have been unable to identify a positive control cell line for endogenous full-length ROS1. The identity of the lower MW band at approximately 60kDa is unknown.

U-118MG cells carry the FIG-ROS1 fusion (PMID : 12661006). Tyrosine phosphorylated ROS1 is downregulated in response to crizotinib treatment (PMID : 24971615)

Target data

Receptor tyrosine kinase (RTK) that plays a role in epithelial cell differentiation and regionalization of the proximal epididymal epithelium. NELL2 is an endogenous ligand for ROS1. Upon endogenous stimulation by NELL2, ROS1 activates the intracellular signaling pathway and triggers epididymal epithelial differentiation and subsequent sperm maturation (By similarity). May activate several downstream signaling pathways related to cell differentiation, proliferation, growth and survival including the PI3 kinase-mTOR signaling pathway. Mediates the phosphorylation of PTPN11, an activator of this pathway. May also phosphorylate and activate the transcription factor STAT3 to control anchorage-independent cell growth. Mediates the phosphorylation and the activation of VAV3, a guanine nucleotide exchange factor regulating cell morphology. May activate other downstream signaling proteins including AKT1, MAPK1, MAPK3, IRS1 and PLCG2.
