RPLP1 also known as the Ribosomal Protein Lateral Stalk Subunit P1 plays an essential mechanical role in protein synthesis within the cell. It is a part of the larger ribosomal 60S subunit and helps in the binding of elongation factors during translation. RPLP1 has a molecular mass of approximately 12 kDa. Its expression is most evident in structures requiring active protein production such as the liver kidney and cardia muscles. Nucleolar localization suggests a role in ribosome biogenesis.
Biological function summary

The protein is involved in regulating ribosome activity for protein synthesis. RPLP1 is a component of the ribosomal stalk which forms an important part of the ribosome complex. It functions with other ribosomal proteins to facilitate the interaction of the ribosome with translational GTPases. The dynamic ability of RPLP1 to dissociate and associate with the ribosome allows the adjustment of translation rates facilitating accurate response to cellular metabolic demands.

Pathways

RPLP1 plays a significant role in the eukaryotic translation pathway including the initiation elongation and termination phases. One of the key interactions is with elongation factors such as EF-2 which helps in the translocation step. This connection highlights RPLP1's role in controlling translational efficiency. Additionally the protein's involvement in the mTOR signaling pathway further emphasizes its importance in cell growth and proliferation relating it to proteins such as mTOR itself and ribosomal proteins like RPL12.

Alterations in RPLP1 expression are associated with certain types of cancer including liver and colorectal cancers. Abnormal RPLP1 activity can disrupt normal protein synthesis rates leading to uncontrolled cell proliferation. This dysfunction interacts with other ribosomal proteins like RPLP2 potentially contributing to tumorigenesis. Furthermore some neurodevelopmental disorders may also link to RPLP1 mutations where it might engage with different proteins involved in neural development.
