Supplementary information

RPN2 also known as Ribophorin II is a subunit of the oligosaccharyltransferase (OST) complex. This protein has a mass of approximately 68 kDa. It is expressed mainly in the rough endoplasmic reticulum playing a role in the N-linked glycosylation of nascent polypeptides. This process is essential for protein folding and stability. RPN2 helps transfer oligosaccharide chains to asparagine residues in nascent proteins.
Biological function summary

RPN2 is integral to the OST complex which consists of multiple subunits each contributing to the glycosylation process. By enabling glycosylation RPN2 ensures proper protein folding allowing proteins to achieve functional conformation and maintain stability. This process affects various cellular activities including protein secretion cell surface receptor expression and immune function.

Pathways

RPN2 plays a significant role in the N-linked glycosylation pathway. This pathway involves the attachment of glycans to proteins influencing their folding distribution and function. RPN2 interacts with other proteins such as STT3A and STT3B which are essential components of the OST complex. This interaction modulates protein modification during synthesis and affects downstream pathways like cell signaling and immune response.

RPN2 has connections to cancer and congenital disorders of glycosylation (CDG). Its altered expression in cancer cells correlates with tumor progression and metastasis where it may impact the function of proteins such as EGFR involved in cell growth and survival. In CDG deficiencies in glycosylation due to RPN2 malfunction can lead to a range of symptoms affecting multiple body systems linking it with several glycosylation-related proteins.

Target data

Subunit of the oligosaccharyl transferase (OST) complex that catalyzes the initial transfer of a defined glycan (Glc(3)Man(9)GlcNAc(2) in eukaryotes) from the lipid carrier dolichol-pyrophosphate to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains, the first step in protein N-glycosylation (PubMed : 31831667). N-glycosylation occurs cotranslationally and the complex associates with the Sec61 complex at the channel-forming translocon complex that mediates protein translocation across the endoplasmic reticulum (ER). All subunits are required for a maximal enzyme activity.
