Supplementary information

RPS19BP1 also known as AROS (Active Regulator Of SIRT1) is a protein with a molecular weight of approximately 14 kDa. It plays an important role in cellular processes. The protein is known to enhance the deacetylase activity of SIRT1 a member of the sirtuin family involved in longevity and metabolism regulation. RPS19BP1/AROS resides mainly in the nucleus and is ubiquitously expressed in various human tissues indicating its broad role in cellular function.
Biological function summary

RPS19BP1/AROS interacts with SIRT1 to modulate deacetylation of target proteins. SIRT1 is an important regulator in various processes such as aging inflammation and cellular stress responses. RPS19BP1 contributes to these processes by activating SIRT1 forming an association to influence cellular outcomes. The interaction between RPS19BP1 and SIRT1 helps in regulating acetylation of non-histone proteins which impacts gene expression and cell survival mechanisms.

Pathways

RPS19BP1/AROS affects cellular pathways including the SIRT1 and AMPK pathways. The SIRT1 pathway is integral to energy balance and stress response and RPS19BP1 modulates this pathway by enhancing SIRT1 activity influencing gluconeogenesis and mitochondrial biogenesis. AMPK is another pathway where RPS19BP1 indirectly plays a role with AMPK known for its role in energy homeostasis. RPS19BP1/AROS through its action on SIRT1 impacts AMPK-related processes thereby linking it to cellular energy regulation and metabolic responses.

RPS19BP1/AROS holds relevance in the context of cancer and metabolic diseases. Enhanced SIRT1 activity through RPS19BP1 association can influence cancer cell survival and proliferation marking it as a potential target for cancer therapies. Additionally metabolic disorders like type 2 diabetes have associations with the dysregulation of pathways involving RPS19BP1 and SIRT1. The interaction with proteins such as p53 which is involved in cell cycle control further illustrates its role in tumorigenesis and metabolic processes.

Target data

Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome. Acts as a chaperone that specifically mediates the integration of RPS19 in state post-A1 (PubMed : 34516797). Direct regulator of SIRT1. Enhances SIRT1-mediated deacetylation of p53/TP53, thereby participating in inhibition of p53/TP53-mediated transcriptional activity (PubMed : 17964266).
