RPS27A also known as Ubiquitin-40S Ribosomal Protein S27a is a fusion protein consisting of ubiquitin and ribosomal protein S27a. It has a molecular mass of approximately 17 kDa. RPS27A is expressed in multiple tissues but has higher expression in actively dividing cells where protein synthesis is high. Mechanically RPS27A contributes to two different functions: ubiquitin marks proteins for degradation while the S27a component incorporates into the 40S ribosomal subunit playing a role in the assembly and function of the ribosome.
Biological function summary

RPS27A serves dual roles due to its hybrid nature. It participates in protein homeostasis through the ubiquitin-proteasome system by tagging proteins for degradation. Additionally it is an essential component of the ribosomal 40S subunit forming part of the cellular machinery that translates mRNA into polypeptides. This dual role allows RPS27A to impact general cell function linking protein turnover with the protein production machinery.

Pathways

RPS27A contributes to the ubiquitin-mediated proteolysis pathway and the ribosome biogenesis pathway. In ubiquitin-mediated proteolysis RPS27A partners with proteins like E1 E2 and E3 enzymes which aid in the transfer and attachment of ubiquitin to target proteins. In ribosome biogenesis RPS27A's ribosomal component joins with other ribosomal proteins within the nucleolus to form the mature ribosome integral to protein synthesis.

RPS27A appears to have associations with cancer and neurodegenerative diseases. In cancer abnormalities in ubiquitin-mediated proteolysis may lead to unchecked cell division implicating RPS27A in tumorigenesis. Additionally disruptions in ribosomal biogenesis to which RPS27A contributes may also play a role in cancer development. Moreover dysfunction in protein degradation mechanisms involving ubiquitin has links to neurodegenerative disorders such as Parkinson’s disease where an accumulation of misfolded or damaged proteins occurs. In such contexts proteins interacting with RPS27A like ubiquitin ligases or proteasomes may be directly involved.

Ubiquitin. Exists either covalently attached to another protein, or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond either as a monomer (monoubiquitin), a polymer linked via different Lys residues of the ubiquitin (polyubiquitin chains) or a linear polymer linked via the initiator Met of the ubiquitin (linear polyubiquitin chains). Polyubiquitin chains, when attached to a target protein, have different functions depending on the Lys residue of the ubiquitin that is linked : Lys-6-linked may be involved in DNA repair; Lys-11-linked is involved in ERAD (endoplasmic reticulum-associated degradation) and in cell-cycle regulation; Lys-29-linked is involved in proteotoxic stress response and cell cycle; Lys-33-linked is involved in kinase modification; Lys-48-linked is involved in protein degradation via the proteasome; Lys-63-linked is involved in endocytosis, DNA-damage responses as well as in signaling processes leading to activation of the transcription factor NF-kappa-B. Linear polymer chains formed via attachment by the initiator Met lead to cell signaling. Ubiquitin is usually conjugated to Lys residues of target proteins, however, in rare cases, conjugation to Cys or Ser residues has been observed. When polyubiquitin is free (unanchored-polyubiquitin), it also has distinct roles, such as in activation of protein kinases, and in signaling.. Small ribosomal subunit protein eS31.
