The blot was stripped and re-blotted for total RPS6 using an affinity purified rabbit anti-RPS6 antibody at 0.04 μg/ml.
Detection : Chemiluminescence with exposure times of 10 seconds (upper panel) or 30 seconds (lower panel).

Lane 1:
HeLa whole cell lysate, grown in serum containing media at 35 µg
Lane 2:
HeLa whole cell lysate, cultured for 24 hours in serum free media; At 24h, serum was replaced for 1 h with 50nM Rapamycin at 35 µg
Lane 3:
HeLa whole cell lysate, cultured for 24 hours in serum free media; At 24h, serum was replaced for 1 h without Rapamycin at 35 µg
Lane 4:
HeLa whole cell lysate, cultured for 24 hours in serum free media; after 24h serum starvation was continued with 50nM Rapamycin at 35 µg
