RREB1 also known as Ras-responsive element-binding protein 1 is a zinc finger transcription factor with a molecular mass of approximately 197 kDa. RREB1 is highly expressed in various tissues including the pancreas lung and prostate. Its primary role involves binding to specific Ras-responsive elements (RREs) in the DNA regulating the transcription of genes that are pivotal for cell differentiation growth and apoptosis.
Biological function summary

The RREB1 protein interacts with other components to activate or repress target gene expression often acting as part of larger transcriptional complexes. It influences key biological processes such as embryonic development and cellular response to stress by controlling genes involved in these pathways. The interconnected function of RREB1 guides essential cellular decisions including how cells adapt to signaling cues in their environment.

Pathways

RREB1 significantly contributes to the Ras signaling pathway and the MAPK/ERK pathway connecting it to important cellular events like proliferation and differentiation. RREB1 functions alongside proteins such as RAS and MEK collaborating to transmit signals from the cell surface to the nucleus. Through these pathways RREB1 plays a role in adjusting cellular responses to external stimuli influencing gene expression patterns critical for diverse cell functions.

The dysregulation of RREB1 has connections to cancer including prostate and pancreatic cancers. Abnormal RREB1 function can lead to uncontrolled cell proliferation due to its role in gene expression regulation. It has interactions with proteins such as KRAS in these disorders contributing to oncogenic transformations. Additionally alterations in RREB1 are linked to diabetes mellitus where its misregulation might affect insulin signaling pathways impacting glucose metabolism and homeostasis.

Target data

Transcription factor that binds specifically to the RAS-responsive elements (RRE) of gene promoters (PubMed : 12700664). Represses the angiotensinogen gene (By similarity). Negatively regulates the transcriptional activity of AR (By similarity). Potentiates the transcriptional activity of NEUROD1 (By similarity). Binds specifically to the allelic variant of the CDKN2A promoter present in Balb/c mice, which leads to a down-regulation of CDKN2A expression in this strain, and, as a consequence, to an elevated susceptibility to pristane-induced tumors (PubMed : 12700664). Promotes brown adipocyte differentiation (PubMed : 27923061). May be involved in Ras/Raf-mediated cell differentiation by enhancing calcitonin expression (By similarity).
