RRP7A also known as Ribosomal RNA Processing 7 Homolog A is a protein involved in the assembly of ribosomal subunits. It has a molecular mass of approximately 33 kDa. RRP7A localizes mainly in the nucleolus where ribosomal RNA (rRNA) synthesis and ribosome assembly occur. It exists in various organisms indicating its conserved function across species. The protein is encoded by the RRP7A gene and its expression is observed in many tissues reflecting its essential function in the basic cellular process of ribosome production.
Biological function summary

RRP7A plays an important role in ribosome biogenesis an essential process for the synthesis of proteins. It is part of the small-subunit (SSU) processome complex which is involved in the maturation of 18S rRNA a critical component of the small ribosomal subunit. The SSU processome is essential for early steps of 40S ribosomal subunit assembly. RRP7A acts in association with other proteins within this complex to facilitate the accurate processing of pre-rRNA into functional rRNA molecules necessary for proper ribosomal function.

Pathways

Ribosomal RNA processing represents a critical aspect of translation and protein synthesis. RRP7A fits into the ribosome biogenesis pathway ensuring the correct formation and function of ribosomes. This pathway involves several other proteins including FBL (Fibrillarin) and DDX5 which work alongside RRP7A within the SSU processome. Together these components contribute to the intricate steps of rRNA processing from transcription to the assembly and transport of ribosomal subunits. This makes RRP7A integral to maintaining cellular protein synthesis capacity.

Impaired function of RRP7A can be related to ribosomopathies and certain cancer types due to disruptions in ribosome production. In some cancers altered ribosome biogenesis results in increased cellular proliferation. RRP7A disruptions link to proteins such as MYC a well-known oncogene that can promote ribosome biogenesis when overexpressed. Understanding RRP7A's role in these conditions can inform studies exploring the therapeutic potential of targeting ribosome biogenesis pathways in disease treatment.

Also plays a role in primary cilia resorption, and cell cycle progression in neurogenesis and neocortex development (PubMed : 33199730). Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome (PubMed : 34516797).
