Supplementary information

RUTBC1 also known as Rab GTPase-activating protein 1 weighs approximately 140 kDa. It serves as a GTPase-activating protein (GAP) to regulate Rab proteins involved in membrane traffic. Most commonly cells express RUTBC1 in brain and muscle tissue mediating processes like vesicle formation and fusion. The gene for RUTBC1 encodes for a protein that features a variety of domains necessary for its function including a RUN domain and a TBC (Tre-2/Bub2/Cdc16) domain.
Biological function summary

Proteins interacting through RUTBC1 regulate processes involving membrane trafficking and cellular dynamics. RUTBC1 often forms part of larger multiprotein complexes that modulate the activity of Rab GTPases. Its expression ensures cells properly direct vesicle transport influencing endocytosis and exocytosis. These processes are important for maintaining cellular homeostasis and signal transduction impacting how cells respond to their external environment.

Pathways

RUTBC1 contributes importantly to the Rab GTPase signaling pathway. By deactivating Rab proteins it affects pathways essential for intracellular membrane trafficking like the endocytic and recycling pathways. RUTBC1 works alongside proteins like Rab5 and Rab7 which are key regulators of early and late endosome dynamics respectively. This collaboration ensures efficient packaging and delivery of cellular materials to proper destinations within a cell.

Research links RUTBC1 to certain neurological disorders and muscle-related diseases. Aberrant functioning of RUTBC1 could disrupt vesicle trafficking leading to conditions like Charcot-Marie-Tooth disease. Associations with neurodegenerative disorders have been noted where RUTBC1 malfunction impacts proteins like TDP-43 contributing to pathological processes. Understanding RUTBC1's interactions and effects could inform therapeutic strategies for these conditions.

Target data

Possesses GTPase activator activity towards RAB32, RAB33B and RAB38 (PubMed : 21808068, PubMed : 26620560). Regulates the trafficking of melanogenic enzymes TYR, TYRP1 and DCT/TYRP2 to melanosomes in melanocytes by inactivating RAB32 and RAB38. Inhibits RAB32 and RAB38 activation both directly by promoting their GTPase activity and indirectly by disrupting the RAB9A-HPS4 interaction which is required for RAB32/38 activation (PubMed : 26620560).
