The 148 kDa zymogen is processed progressively into two membrane-bound 120 and 106 kDa forms in the endoplasmic reticulum, and late into a secreted 98 kDa form. The propeptide is autocatalytically removed through an intramolecular cleavage after Leu-186. Further cleavage generates 14, 10, and 8 kDa intermediates.

Sphingosine-1-phosphate (S1P) also known as SPC is a signaling sphingolipid with a molecular mass of approximately 379 Da. It plays an important role in cellular signaling by binding to a family of G-protein-coupled receptors known as S1P receptors (S1PR1-5). S1P is expressed in various tissues including the immune system cardiovascular system and central nervous system. It exhibits a strong influence on cell survival proliferation and migration through its interactions with these receptors.
Biological function summary

S1P regulates immune cell trafficking vascular maturation and endothelial cell integrity. It acts in concert with other bioactive lipids as part of larger signaling complexes. This signaling molecule influences the barrier function of endothelial cells and it exerts control over lymphocyte egress from secondary lymphoid organs. S1P is also implicated in the stabilization of endothelial adherens junctions further highlighting its role in maintaining vascular homeostasis.

Pathways

The S1P signaling contributes significantly to the sphingolipid metabolism pathway and the MAPK signaling pathway. In the sphingolipid pathway S1P is synthesized from sphingosine by sphingosine kinases and subsequently acts through its receptors to exert various cellular effects. The MAPK signaling pathway involves proteins like ERK where S1P can influence cellular responses like proliferation and survival. This tight regulation of signaling pathways secures S1P's role as a central lipid mediator in processes such as inflammation and angiogenesis.

S1P has associations with multiple sclerosis and cancer. In multiple sclerosis S1P modulation affects lymphocyte circulation with its receptor S1PR1 being an important player in the disease's treatment approach by controlling lymphocyte migration. In cancer S1P signaling can promote tumor growth and metastasis linked to proteins like VEGF which enhance angiogenesis. Understanding S1P's complex interactions in these diseases enables the development of therapeutic targets to mitigate pathological conditions.
