Immunohistochemical analysis of paraffin-embedded Rat cerebrum tissue labeling S1P1/EDG1 with ab259902 at 1/100 (5.08 ug/ml) followed by a LeicaDS9800 (Bond™ Polymer Refine Detection) at Ready to use dilution. Cytoplasmic staining in rat cerebrum.The section was incubated with ab254360 for 30 minutes at room temperature.The immunostaining was performed on a Leica Biosystems BOND® RX instrument Counterstained with Hematoxylin.
Secondary antibody only control : Secondary antibody is LeicaDS9800 (Bond™ Polymer Refine Detection) at Ready to use dilution.
Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 minutes.

S1P1/EDG1 was immunoprecipitated from 0.35 mg Mouse brain tissue lysate 10 ug with ab259902 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab259902 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP) (ab131366) was used at 1/5000 dilution.

Biological function summary
S1P1 is involved in the regulation of immune cell trafficking and vascular stability. As a part of a receptor complex it activates intracellular signaling cascades that influence cell migration and vascular maturation. By binding to S1P S1P1 controls the egress of lymphocytes from lymphoid tissues indicating its important role in immune surveillance and response.
Pathways
S1P1 serves as an important component in the S1P signaling pathway and the phosphoinositide 3-kinase (PI3K) pathway. In the S1P pathway it regulates endothelial cell barrier function and vascular maturation. In the PI3K pathway S1P1 influences cell survival and migration which relates it to protein kinases like Akt and other signaling molecules that further modify cellular responses.
