Supplementary information

SAC3D1 also known as SH3- and SAC domain-containing protein 1 is a protein that plays a mechanical role in intracellular transport and phosphoinositide metabolism. The protein has a mass of approximately 94 kDa and is expressed in various tissues with higher levels in the brain and testis. SAC3D1 contains an SH3 domain and a phosphatase domain which are important for its function in cellular processes.
Biological function summary

SAC3D1 interacts with other proteins to form a complex critical for phosphatidylinositol-3-phosphate binding and turnover. This activity is essential in membrane trafficking regulating endosomal dynamics and impacting cellular signaling pathways. SAC3D1's ability to mediate phosphoinositide signaling suggests that it plays a role in cell growth and differentiation maintaining normal cellular functions.

Pathways

SAC3D1 is involved in the phosphoinositide signaling pathway and the endocytic recycling pathway. These pathways include interactions with proteins like MTMR2 and FIG4 which are important for lipid phosphatase activity and vesicle recycling. SAC3D1's association with these proteins highlights its influence on maintaining endosome homeostasis within cells important for proper signaling and trafficking functions.

SAC3D1 has been linked to conditions such as Charcot-Marie-Tooth disease and Amyotrophic Lateral Sclerosis (ALS). Its interactions with the FIG4 protein is significant as mutations in FIG4 relate to these disorders. Disruptions in SAC3D1 function can lead to defects in lipid metabolism and neuronal maintenance contributing to the pathogenesis of these neurodegenerative diseases.
