SACM1L also known as Sac1 is an integral enzyme in the cell performing its role as a phosphoinositide phosphatase. It has a mass of approximately 59 kDa. SACM1L localizes in the cytoplasm and the membrane systems of the endoplasmic reticulum and Golgi apparatus where it performs its enzymatic activities. Its expression is observed in many tissues with particular abundance in the brain and testis. The protein's ability to dephosphorylate phosphoinositides highlights its mechanical role in cellular lipid metabolism.
Biological function summary

This enzyme regulates intracellular lipid signaling and trafficking by modulating the levels of phosphoinositides. SACM1L is part of a protein complex involving other regulatory proteins that maintain the balance of lipid phosphates. Through its enzymatic activity SACM1L influences membrane dynamics and vesicular transport within cells. Taken together these activities show its importance in maintaining cellular homeostasis and function.

Phosphoinositide phosphatase which catalyzes the hydrolysis of phosphatidylinositol 4-phosphate (PtdIns(4)P) (PubMed : 24209621, PubMed : 27044890, PubMed : 29461204, PubMed : 30659099). Can also catalyze the hydrolysis of phosphatidylinositol 3-phosphate (PtdIns(3)P) and has low activity towards phosphatidylinositol-3,5-bisphosphate (PtdIns(3,5)P2) (By similarity). Shows a very robust PtdIns(4)P phosphatase activity when it binds PtdIns(4)P in a 'cis' configuration in the cellular environment, with much less activity seen when it binds PtdIns(4)P in 'trans' configuration (PubMed : 24209621, PubMed : 29461204, PubMed : 30659099). PtdIns(4)P phosphatase activity (when it binds PtdIns(4)P in 'trans' configuration) is enhanced in the presence of PLEKHA3 (PubMed : 30659099).
