Binds to scaffold/matrix attachment region (S/MAR) DNA and forms a molecular assembly point to allow the formation of a 'transcriptosomal' complex (consisting of SR proteins and RNA polymerase II) coupling transcription and RNA processing (PubMed:9671816). Functions as an estrogen receptor corepressor and can also bind to the HSP27 promoter and decrease its transcription (PubMed:12660241). Thereby acts as a negative regulator of cell proliferation (PubMed:12660241). When associated with RBMX, binds to and stimulates transcription from the SREBF1 promoter (By similarity).

SAFB also known as Scaffold Attachment Factor B is a protein that participates in multiple cellular functions. It has a molecular weight of approximately 135 kDa. Researchers observe its expression in the nucleus of a variety of cell types including epithelial and fibroblast cells. In a mechanical context SAFB plays a role in binding to scaffold/matrix-attachment regions (S/MARs) of DNA influencing transcription and chromatin organization.
SAFB works as a multifunctional protein that regulates gene expression and RNA processing. It acts as part of protein complexes such as those involved in modulating estrogen receptor activity. SAFB interacts with chromatin and RNA acting as a bridge between these important cellular components. It influences transcription by functioning as both a transcriptional repressor and activator depending on the context within the cell.
Several cellular interactions highlight the role of SAFB within nuclear signaling networks like the estrogen signaling pathway and stress response pathways.
