Sarcalumenin is a calcium-binding protein with a molecular weight of approximately 160 kDa. It's also known as SRL. This protein is expressed mainly in the skeletal and cardiac muscle tissues where it populates the sarcoplasmic reticulum. Sarcalumenin plays a role in modulating calcium dynamics by storing and buffering calcium ions which is critical for muscle contraction and relaxation processes.
Biological function summary

This calcium-binding protein is involved in various muscle functions by being part of the sarcoplasmic reticulum membrane complex. Sarcalumenin assists in the regulation of calcium uptake and release essential for proper muscle function. It interacts with other proteins such as calsequestrin and triadin contributing to efficient calcium cycling which is necessary for rhythmic contractions of cardiac and skeletal muscles.

Pathways

Sarcalumenin plays an integral part in the calcium signaling pathway which is fundamental for muscle contraction and energy metabolism. It works closely with proteins like SERCA (Sarcoplasmic/Endoplasmic Reticulum Ca²⁺ ATPase) to facilitate the cycling of calcium ions across the sarcoplasmic reticulum. Furthermore it contributes to the excitation-contraction coupling pathway where its interaction with calsequestrin ensures synchronized muscle activity.

Altered expression or mutation of sarcalumenin has associations with cardiac-related conditions such as arrhythmias and cardiomyopathy. The protein's interaction with RyR2 (ryanodine receptor 2) highlights its role in these disorders. In skeletal muscles abnormalities in sarcalumenin function can lead to muscle fatigue and weakened contraction sometimes observed in myopathy patients. The balance and interaction with calsequestrin also suggest its involvement in maintaining muscle health.
