Immunofluorescent analysis of human skeletal muscle myoblast cells fixed using 4% formaldehyde (reconstituted in 1X PBS) for 10 min at room temperature and permeabilized using 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling endogenous sarcomeric alpha actin with ab313448 at 2 ?g/mL followed by Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor 488 conjugate at 1/2000 dilution. Panel a) shows representative cells that were stained for detection and localization of sarcomeric alpha actin (green), Panel b) is stained for nuclei (blue) using DAPI. Panel c) represents cytoskeletal F-actin staining using Rhodamine Phalloidin at 1/300 dilution. Panel d) is a composite image of Panels a, b and c clearly demonstrating cytoskeletal localization of sarcomeric alpha actin. Sarcomeric Alpha Actinin often referred to as alpha-actinin or A-actinin is a protein that functions mechanically as a cross-linker of actin filaments in the sarcomere. It plays a critical role in organizing the actin cytoskeleton providing structural integrity and facilitating muscle contraction. The protein typically has a molecular mass of approximately 100 kDa. It is expressed in a variety of tissues including skeletal and cardiac muscle where it establishes the architecture of the Z-disc by anchoring actin filaments.
