SARS-CoV-2 nsp12 also known as nonstructural protein 12 functions as an RNA-dependent RNA polymerase (RdRp) important for viral RNA genome replication. This protein weighs approximately 106 kilodaltons and derives from the SARS-CoV-2 pathogen the virus responsible for COVID-19. Nsp12 integral to the replication complex is expressed primarily in the host cell's cytoplasm during coronavirus infection. Together with other nonstructural proteins it directs the replication and transcription of the viral genome thereby playing an important role in the viral life cycle.
Nonstructural protein 12 associates with cofactors nsp7 and nsp8 to form a functional polymerase complex essential for replicating the viral RNA genome. This complex efficiently extends RNA strands from RNA templates using the 3' end as a starting point. Nsp12 contains a conserved polymerase domain which ensures the fidelity of replication. It performs extensive proofreading alongside overlapping proteins like nsp14 which provide exonuclease activity to correct errors sustaining the propagation of the virus within the host.
The nsp12 protein directly interacts with mechanisms like viral RNA replication and viral mRNA synthesis. It holds a central position in the coronavirus replication-transcription complex working closely with other nonstructural proteins such as nsp7 and nsp8. These interactions enable the synthesis of full-length genomic RNA and shorter subgenomic RNAs critical for translating viral structural proteins therefore facilitating the continuous cycle of infection and propagation.
Nsp12's activity directly relates to COVID-19 pathogenesis making it a significant target for antiviral treatments. The inhibition of nsp12 through therapeutic agents can effectively disrupt viral RNA synthesis curbing the replication of SARS-CoV-2. Remdesivir a nucleotide analog drug that targets the RdRp activity of nsp12 exemplifies this strategy specifically impeding the SARS-CoV-2 replication process. Additionally variations in nsp12 and its associated protein interactions influence the severity and progression of coronavirus infections in affected individuals.

For immunofluorescence analysis, mock and transfected HEK293T cells were fixed in 4% paraformaldehyde at room temperature for 15 minutes and permeabilized for detection of SARS CoV 2 COVID 19 nsp12 RdRP using ab308372 at 1/500 dilution (green). Blue: Fluoroshield with DAPI.
Non-transfected (–) and transfected (+) 293T whole cell extracts (30 μg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with SARS-CoV-2 (COVID-19) nsp12 / RdRP antibody [HL1444] (ab308372) diluted at 1/5000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.
