Non-transfected (–) and transfected (+) 293T whole cell extracts (30 ug) were separated by 12% SDS-PAGE and the membrane was blotted with ab317206 diluted at 1 : 1000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

Biological function summary
Nsp3 is critical in the formation of the replication complex where it collaborates with other nonstructural proteins like nsp4 and nsp6. These interactions facilitate the modification of host cell membranes creating a conducive environment for viral RNA synthesis. In addition nsp3's enzymatic activity is necessary for inhibiting host immune responses supporting the virus's ability to establish infection within a host.
Pathways
Nsp3 participates in the viral life cycle by influencing key pathways including the autophagy pathway and translation initiation. Nsp3 can modulate the host's autophagy which typically helps in degrading cellular components to favor viral replication. Additionally nsp3 interferes with translation initiation affecting the host's ability to synthesize proteins therefore aiding in the initiation of viral protein synthesis. It cooperates with proteins like nsp5 and nsp12 in these pathways further ensuring efficient production of viral components.
