Intracellular Flow Cytometry analysis of HEK-293T (Human embryonic kidney epithelial cell) cells transfected with a SARS-CoV-2 nsp9 (Left) or SARS-CoV2 nsp1 (Right) expression vector containing a myc tag labeling SARS-COV-2 nsp1 with ab284631 at 1/5000 dilution (left and right). Cells were fixed with 4% Paraformaldehyde and permeabilised with 90% Methanol. A Goat Anti-Rabbit IgG H&L (Alexa Fluor® 647, ab150083) secondary antibody was used at 1/5000.

Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HEK-293T (Human epithelial cell line from embryonic kidney transformed with large T antigen) cells labelling SARS-COV-2 nsp1 with primary antibody anti-SARS-COV-2 nsp1 (ab284631) at 1/100 dilution, followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150081) secondary antibody at 1/1000 dilution. Confocal image showing positive staining in HEK-293T cells transfected with a SARS-CoV2 nsp1 expression vector containing a myc tag, no staining in HEK-293T cells transfected with a SARS-CoV2 nsp9 expression vector containing a myc tag. Myc-Tag Mouse mab (Alexa Fluor® 647) (red) was used as a counterstain at 1/100 dilution. The nuclear counter stain is DAPI (blue).

ab284631 was shown to bind specifically to SARS-CoV-2 NSP1 protein in Western blot.

Samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged.

Secondary antibodies used were Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

Blocking buffer : 3% milk in TBS-0.1% Tween® 20 (TBS-T)

SARS-CoV-2 nsp1 also known as non-structural protein 1 is a viral protein with a molecular mass of around 19.8 kDa. It is produced by the SARS-CoV-2 virus the agent responsible for the COVID-19 pandemic. Nsp1 is expressed in the cytoplasm of infected host cells. It is important for the virus as it plays a significant role in the regulation of host cell processes making it an interesting target for antiviral research.
Biological function summary

This non-structural protein inhibits host cellular gene expression. Nsp1 disrupts the host's mRNA translation machinery by directly interacting with the ribosomal 40S subunit. This interference enables the virus to suppress the cell's immune response aiding viral replication and assembly. Nsp1 does not form part of a large complex but functions independently to achieve its suppressive effects.

Pathways

Nsp1's function aligns with pathways related to host translation processes and immune evasion tactics. It acts to inhibit the IFN-mediated signaling pathway preventing the usual anti-viral response of the cell.
