This data was developed using the same antibody clone in a different buffer formulation (ab286911).
Rabbit monoclonal to SARS-CoV2 orf6 (ab286911) staining at 1/1000 dilution, shown in Green; Mouse anti-GAPDH antibody [6C5] (ab8245) at 1/20000 dilution, shown in red.
In Western blot, ab286911 was shown to bind specifically to SARS-CoV2 ORF6 protein. A band was observed at 13 kDa in HEK-293T transfected with SARS-CoV-2 orf6 cell lysates with no signal observed at this size in HEK-293T transfected with SARS-CoV-1 orf6 cell lysates.
Samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4°C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged.
Secondary antibodies used were Goat anti-Rabbit IgG H&L 800CW and Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution.

Biological function summary
The SARS2-CoV-2 ORF6 protein interacts with host cell proteins to inhibit the host's immune defense. It is known to inhibit the nuclear import of host cell transcription factors like STAT1 and STAT2 effectively dampening the interferon-mediated antiviral response. The protein does not function alone but interacts with other viral components to optimize the virus's evasion strategies.
Pathways
The SARS2-CoV-2 ORF6 protein is involved in the interferon signaling pathway. By impairing this pathway the protein hinders the expression of interferon-stimulated genes that are important for antiviral defense. ORF6 has interactions with host proteins involved in these pathways contributing to an effective immune evasion strategy comparable to other SARS-CoV-2 proteins like NSP1.
