Western blot - Anti-SARS2-CoV-2 ORF6 antibody [EPR24849-21] - BSA and Azide free (AB286919)

This data was developed using the same antibody clone in a different buffer formulation (ab286911).

Rabbit monoclonal to SARS-CoV2 orf6 (ab286911) staining at 1/1000 dilution, shown in Green; Mouse anti-GAPDH antibody [6C5] (ab8245) at 1/20000 dilution, shown in red.

In Western blot, ab286911 was shown to bind specifically to SARS-CoV2 ORF6 protein. A band was observed at 13 kDa in HEK-293T transfected with SARS-CoV-2 orf6 cell lysates with no signal observed at this size in HEK-293T transfected with SARS-CoV-1 orf6 cell lysates.

Samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4°C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged.

Secondary antibodies used were Goat anti-Rabbit IgG H&L 800CW and Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution.

Product details

This is the carrier free version of ab286911.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

SARS2-CoV-2 ORF6 also known as ORF6 protein is a small accessory protein of the SARS-CoV-2 virus with a molecular mass around 7 kDa. ORF6 is non-structural and plays an important role in the viral lifecycle. It is expressed in infected host cells alongside other viral proteins. ORF6 has a significant role in manipulating the host's immune response to ensure successful viral replication and spread within the host organism.
Biological function summary

The SARS2-CoV-2 ORF6 protein interacts with host cell proteins to inhibit the host's immune defense. It is known to inhibit the nuclear import of host cell transcription factors like STAT1 and STAT2 effectively dampening the interferon-mediated antiviral response. The protein does not function alone but interacts with other viral components to optimize the virus's evasion strategies.

Pathways

The SARS2-CoV-2 ORF6 protein is involved in the interferon signaling pathway. By impairing this pathway the protein hinders the expression of interferon-stimulated genes that are important for antiviral defense. ORF6 has interactions with host proteins involved in these pathways contributing to an effective immune evasion strategy comparable to other SARS-CoV-2 proteins like NSP1.

The activity of SARS2-CoV-2 ORF6 contributes significantly to the pathogenesis of COVID-19 the disease caused by the SARS-CoV-2 virus. Its function in inhibiting the immune response may lead to severe respiratory symptoms due to uncontrolled viral replication. Connections between ORF6 and other viral proteins like ORF3a have shown a cumulative effect on disease severity highlighting its role in COVID-19 pathology.

Target data

Disrupts bidirectional nucleocytoplasmic transport by interacting with the host RAE1-NUP98 complex (PubMed : 33360543, PubMed : 33849972). Disrupts cell nuclear import complex formation by tethering karyopherin alpha 2 and karyopherin beta 1 to the membrane (PubMed : 32979938). Retention of import factors at the ER/Golgi membrane leads to a loss of transport into the nucleus (By similarity). Prevents STAT1 nuclear translocation in response to interferon signaling, thus blocking the expression of interferon stimulated genes (ISGs) that display multiple antiviral activities (PubMed : 33097660). Suppresses IFN-beta production possibly by blocking IRF3 nuclear translocation (PubMed : 32979938). Might induce accumulation of host HNRNPA1 (PubMed : 33360543).. May play a role in viral double membrane vesicles networks to enhance viral replication.
