SASS6 also known as spindle assembly 6 homolog is a protein that plays an important role in centriole biogenesis. It has a molecular mass of approximately 82 kDa. SASS6 expresses not only in dividing cells but also in tissues where active cell division takes place including the brain testis and other proliferative tissues. Its distribution indicates its involvement in cellular processes that require precise organelle replication and segregation.
Biological function summary

SASS6 integrates into a complex essential for centriole duplication which is fundamental for accurate mitotic spindle formation. It forms a structural core in the assembling centrioles ensuring their cylindrical shape. The protein helps in organizing microtubule triplets around a central hub which is vital for proper cell division and maintenance of genomic stability.

Pathways

The protein contributes significantly to the cell cycle regulation and centrosome cohesion. It participates in the centriole duplication pathway which interacts with other proteins like STIL and CEP135. Through this interaction SASS6 ensures efficient progression and regulation within the centrosome cycle which is critical for cell cycle checkpoints and prevention of aneuploidy.

Mutations or dysregulation of SASS6 are linked to microcephaly and other developmental anomalies. Such conditions often result from improper centriole formation impacting neurogenesis and brain size. The involvement of SASS6 also cross-references with proteins such as CEP63 in these disorders highlighting a shared pathway impacting centrosome-related abnormalities.

Required for centrosome biogenesis and duplication : required both for mother-centriole-dependent centriole duplication and deuterosome-dependent centriole amplification in multiciliated cells (PubMed : 15665853, PubMed : 16244668, PubMed : 17681131). Not required for centriole formation in embryonic stem cells but necessary to maintain centriole architecture (By similarity).
