SCAMP3 also known as Secretory Carrier-Associated Membrane Protein 3 weighs approximately 38 kDa. It is a member of the SCAMP family characterized by several cytoplasmic domains and transmembrane regions. SCAMP3 is expressed in various tissues with notable expression in kidney lung and brain tissue. Mechanically it participates in membrane trafficking within cells particularly involved in processes of endocytosis and recycling of membrane proteins.
Biological function summary

SCAMP3 plays a role in the trafficking of proteins between cell compartments. It is part of the protein complex that facilitates efficient recycling of receptors and other proteins from endosomes back to the plasma membrane or towards lysosomal degradation pathways. SCAMP3 influences the endocytic pathway efficiency and impacts cellular activities such as signal transduction and nutrient intake.

Pathways

SCAMP3 functions in the endosomal trafficking pathway and is associated with the epidermal growth factor receptor (EGFR) signaling pathway. It works alongside proteins like Eps15 and AP-2 adapter protein to help modulate the sorting and recycling of endocytosed proteins. SCAMP3 is critical in determining the routing of internalized cargo which impacts signaling pathway duration and intensity.

SCAMP3 has been linked to neurodegenerative disorders and certain types of cancer. Altered SCAMP3 expression can interfere with protein trafficking contributing to pathological conditions like Alzheimer’s disease. Furthermore its relationship with EGFR ties SCAMP3 to oncogenic processes where dysregulation is often seen in lung cancer. Proteins such as EGFR and amyloid precursor protein (APP) exhibit pathological connections with SCAMP3 in these conditions.
