The SCN4A gene encodes the Nav1.4 protein also known as the sodium channel voltage-gated type IV alpha subunit. Nav1.4 is a voltage-gated sodium channel with a molecular weight around 250 kDa. It mainly facilitates the rapid influx of sodium ions into skeletal muscle cells which is important for the initiation and conduction of action potentials. Expression of Nav1.4 is strong in skeletal muscle tissues playing an important role in muscle excitability and contraction. It serves as a pivotal point in the regulation of skeletal muscle function.
Biological function summary

Nav1.4 is essential in generating and propagating action potentials necessary for muscle contraction. This protein functions as part of a complex that includes beta subunits that help modulate its properties. The precise gating and ion permeability of Nav1.4 are important for proper muscle responsiveness. Pore-forming subunit of Nav1.4, a voltage-gated sodium (Nav) channel that directly mediates the depolarizing phase of action potentials in excitable membranes. Navs, also called VGSCs (voltage-gated sodium channels) or VDSCs (voltage-dependent sodium channels), operate by switching between closed and open conformations depending on the voltage difference across the membrane. In the open conformation they allow Na(+) ions to selectively pass through the pore, along their electrochemical gradient. The influx of Na+ ions provokes membrane depolarization, initiating the propagation of electrical signals throughout cells and tissues (PubMed : 11834499). Highly expressed in skeletal muscles, Nav1.4 generates the action potential crucial for muscle contraction (PubMed : 18317596, PubMed : 21881211).
