SCRT1 also known as Scratch family transcriptional repressor 1 is a zinc finger transcription factor with a molecular weight of approximately 40 kDa. It is expressed in the central nervous system particularly in areas associated with neuronal development and differentiation. The SCRT1 protein binds to specific DNA sequences modulating gene expression by repressing target genes involved in cellular processes.
Biological function summary

SCRT1 plays a role in regulating neuronal differentiation and migration during development. It is not part of a large protein complex but interacts with other transcription factors to modify transcriptional activities. SCRT1 achieves its biological function by repressing genes essential for the maintenance of neural progenitors thereby facilitating the transition to mature neuronal phenotypes.

Pathways

SCRT1 influences neural development pathways interacting with Notch signaling and other developmental pathways. Within these frameworks SCRT1 negatively regulates the expression of genes downstream of proteins like Hes1 and Ascl1 which are important for maintaining neuroprogenitor characteristics and promoting neuronal maturation. This tight regulation ensures proper neuronal development by controlling the balance between neural cell proliferation and differentiation.

Mutations or dysregulation of SCRT1 have connections to neurodevelopmental disorders such as intellectual disabilities and autism spectrum disorders. Additionally SCRT1 interacts with proteins like REST which is implicated in the pathogenesis of such conditions. Altered SCRT1 activity may contribute to the failure of neuronal cells to correctly differentiate leading to the manifestations seen in these disorders.
