Is a positive regulator of the BMP signaling pathway, required for proper chondrogenesis, osteogenesis and skeletal development. It acts as a coreceptor for BMP ligands, particularly BMP2 and BMP4, facilitating their interactions with BMP type I receptors (PubMed:33308444). It is required for ligand-induced recruitment of BMP receptors to lipid rafts (By similarity). Binds to TGFBR2 and activates TGFB signaling. In lung cancer cells, could serve as an endogenous autocrine and paracrine ligand of TGFBR2, which could regulate TGFBR2 signaling and hence modulate epithelial-mesenchymal transition and cancer progression.

The protein SCUBE3 also known as Signal Peptide CUB domain and Epidermal Growth Factor-like domain-containing protein 3 is a member of the SCUBE family. It is characterized by its mass of approximately 110 kDa. SCUBE3 is primarily expressed in various tissues including kidney lung heart and bone. Structurally it has important domains such as CUB and EGF-like domains that assist in its interaction with different molecules in the extracellular environment.
SCUBE3 functions as a coreceptor playing a significant role in modulating developmental and morphogenetic processes. It is part of a complex involved in cell signaling enhancing interactions between growth factors and their receptors. Through these interactions SCUBE3 regulates cell proliferation differentiation and movement essential in embryonic development and tissue homeostasis.
SCUBE3 plays a role in critical signaling pathways such as the Hedgehog and transforming growth factor-beta (TGF-β) pathways. It interacts with proteins like Smoothened (Smo) in the Hedgehog pathway and TGF-β receptors influencing cellular responses. By modulating these pathways SCUBE3 contributes to signaling cascades that are important for organ development and tissue repair.
SCUBE3 relates to pulmonary fibrosis and cancer. It has a connection with proteins like fibroblast growth factor (FGF) in these conditions. In pulmonary fibrosis aberrant SCUBE3 activity influences fibroblast proliferation and extracellular matrix deposition. In cancer particularly in aggressive tumor types SCUBE3's role in enhancing growth factor signaling can contribute to uncontrolled cell growth and metastasis.
