SCXA also known as Scleraxis A is a transcription factor with a molecular mass of approximately 23 kDa. This protein plays a significant mechanical role in gene expression regulation particularly in connective tissues where it binds to specific DNA sequences. SCXA expression occurs in tendons ligaments and other fibrous tissues where it serves an essential function in development and homeostasis.
Biological function summary

The Scleraxis A transcription factor regulates the differentiation of progenitor cells into tenocytes the cells that form tendons. SCXA operates within a transcriptional complex that directs the expression of genes necessary for tendon extracellular matrix synthesis and organization. The regulation by SCXA ensures the structural integrity and function of tendons and connective tissues critical for movement and force transmission.

Pathways

SCXA participates in key developmental signaling processes including the TGF-beta signaling pathway and the Notch signaling pathway. These pathways influence various biological processes related to cellular differentiation and tissue development. SCXA interacts with other proteins such as Mohawk homeobox (MKX) which also functions in tendon development modulating the cellular responses through these pathways to impact the differentiation and maturation of connective tissue cells.

Aberrant SCXA expression or function links to tendon-related pathologies and fibrotic conditions. Research shows potential connections between SCXA and conditions like tendinopathy characterized by tendon degeneration and impaired healing. SCXA also interacts with proteins such as scleraxis B (SCXB) in the pathological context where disruptions in their activity may lead to changes in tendon structure and mechanics impacting tissue response to stress and injury.
