This data was developed using ab307722, the same antibody clone in a different buffer formulation. Immunohistochemical analysis of paraffin-embedded Mouse cerebrum tissue labeling SCXA with ab307722 at 1/500 (0.908 ug/ml) followed by a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection). Negative control : No staining on adult mouse cerebrum.The section was incubated with ab307722 for 30 mins at room temperature.The immunostaining was performed on a Leica Biosystems BOND® RX instrument Counterstained with Hematoxylin. Secondary antibody only control : Secondary antibody is a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection). Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins

Biological function summary
The Scleraxis A transcription factor regulates the differentiation of progenitor cells into tenocytes the cells that form tendons. SCXA operates within a transcriptional complex that directs the expression of genes necessary for tendon extracellular matrix synthesis and organization. The regulation by SCXA ensures the structural integrity and function of tendons and connective tissues critical for movement and force transmission.
Pathways
SCXA participates in key developmental signaling processes including the TGF-beta signaling pathway and the Notch signaling pathway. These pathways influence various biological processes related to cellular differentiation and tissue development. SCXA interacts with other proteins such as Mohawk homeobox (MKX) which also functions in tendon development modulating the cellular responses through these pathways to impact the differentiation and maturation of connective tissue cells.
