Component of the coat protein complex II (COPII) which promotes the formation of transport vesicles from the endoplasmic reticulum (ER). The coat has two main functions, the physical deformation of the endoplasmic reticulum membrane into vesicles and the selection of cargo molecules for their transport to the Golgi complex (PubMed:17499046, PubMed:18843296, PubMed:20427317). Plays a central role in cargo selection within the COPII complex and together with SEC24C may have a different specificity compared to SEC24A and SEC24B (PubMed:17499046, PubMed:18843296, PubMed:20427317). May more specifically package GPI-anchored proteins through the cargo receptor TMED10 (PubMed:20427317). May also be specific for IxM motif-containing cargos like the SNAREs GOSR2 and STX5 (PubMed:18843296).

SEC24D also known as SEC24-related protein D plays an important role in the transport of secretory proteins from the endoplasmic reticulum to the Golgi apparatus by mediating cargo selection for vesicle budding. This protein is part of the COPII coat complex. SEC24D has a molecular mass of approximately 130 kDa. It is widely expressed in most tissues with higher expression levels observed in the liver and testes. Its function in vesicular trafficking makes it essential for maintaining cellular protein transport.
The protein is involved in the packaging of cargo into COPII vesicles. It operates within the COPII complex which includes proteins like SEC23 SEC13 SEC31 and SAR1. SEC24D functions by recognizing and binding specific cargo sorting signals ensuring proteins reach their correct destination. This process is fundamental in maintaining proper cellular organization and function by facilitating efficient secretory pathways.
The SEC24D protein is important in the secretory pathway one of two key pathways for protein transport. It collaborates with the COPII machinery linking endoplasmic reticulum export to various cellular functions. SEC24D works closely with SEC23 in forming budding vesicles which eventually fuse with the Golgi apparatus. The COPII pathway is tightly connected with the ER stress response pathway ensuring cellular adaptation to changes in protein-folding demands.
SEC24D mutations have been linked to congenital disorders of glycosylation and skeletal abnormalities such as osteogenesis imperfecta. SEC24D interacts with other components of the COPII complex like SEC23 which also play a role in these conditions. Understanding the function of SEC24D can provide insights into therapeutic strategies aimed at correcting dysregulated protein trafficking pathways associated with such disorders.
