The SEC63 protein also known as NNF1 plays a critical role in the process of protein translocation across the endoplasmic reticulum (ER) membrane. It has a molecular mass of approximately 80 kDa. SEC63 is widely expressed in various tissues with notable expression in the ER where it functions as part of the translocon complex facilitating the proper insertion and folding of nascent polypeptides into the ER. This protein contains several distinct domains that contribute to its function in protein trafficking and biogenesis within the cell.
Biological function summary

SEC63 participates in cooperation with other components of the Sec61 translocon complex to assist in the translocation of polypeptides from the cytosol into the ER. It is essential for the proper assembly of membrane and secretory proteins directly impacting protein maturation. SEC63 interacts with proteins such as SEC62 and BIP which further stabilize the translocon and assist in the unfolding and refolding of polypeptides. Through this complex SEC63 plays an integral role in maintaining ER homeostasis.

Pathways

SEC63 is an integral component of the protein secretion pathway and the ER-associated degradation (ERAD) pathway. In the protein secretion pathway SEC63 cooperates with proteins like SEC61 to facilitate the proper folding and assembly of proteins destined for secretion. In the ERAD pathway SEC63 aids in recognizing and targeting misfolded proteins for degradation thereby preventing accumulation of defective proteins. By participating in these pathways SEC63 ensures the fidelity of protein quality control and homeostasis within the cellular environment.

Mutations in SEC63 have been linked to polycystic liver disease (PCLD) and can also influence processes related to cancer development. In PCLD the SEC63 protein's impaired function leads to defective protein processing contributing to cyst formation in liver tissue. SEC63's involvement in cancer relates to its role in maintaining protein quality control where disruption can result in an environment conducive to tumor growth. The interaction between SEC63 and proteins such as polycystin-1 and polycystin-2 highlights its importance in these pathological conditions as these proteins are similarly implicated in such diseases.
