Secretory lectin ZG16 also known as ZG16p (zymogen granule protein 16) plays an important role in the cell by binding carbohydrates. This protein has a molecular mass of approximately 16 kDa and is involved in glycan recognition showcasing its affinity for mannose. ZG16 is expressed mainly in the pancreas notably in the zymogen granules of acinar cells where it contributes to the packaging of secretory products.
Biological function summary

This lectin-based protein maintains the integrity and function of zymogen granules. ZG16 interacts with glycosaminoglycans facilitating the trafficking and localization of these granules within the cell. It is not known to be part of a larger protein complex but might interact transiently with other components of the zymogen granules. This molecular activity assists in efficient enzyme storage and secretion fundamental for normal pancreatic function.

Pathways

ZG16 plays a role in the cellular processes of exocytosis and protein trafficking. Its function aligns with the MAPK signaling pathway as well as the vesicle-mediated transport pathway. In these pathways ZG16's interaction with proteins like MIST1 and RabGTPase influences granule maturation and secretory pathways which are essential for the controlled release of digestive enzymes.

ZG16 links to conditions such as pancreatitis and cystic fibrosis where proper enzyme secretion is compromised. During pancreatitis inflammation affects ZG16 expression altering granule dynamics and impacting digestion. In cystic fibrosis although the primary defect lies in CFTR changes in ZG16 expression may contribute to malabsorption issues due to improper enzyme activation. Understanding the role of ZG16 in these diseases provides insights into potential therapeutic avenues targeting these proteins.
