Immunoprecipitation - Anti-SEMA4A antibody [EPR27097-20] (AB314648)
SEMA4A was immunoprecipitated from 0.35 mg THP-1(human monocytic leukemia monocyte) membrane fraction with ab314648 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab314648 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution.
Lane 1 : THP-1(human monocytic leukemia monocyte) membrane fraction
Lane 2 : ab314648 IP in THP-1(human monocytic leukemia monocyte) membrane fraction
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab314648 in THP-1 membrane fraction
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
All lanes:

Biological function summary
The interaction of SEMA4A with its receptors such as Plexin B1 and TIM-2 mediates important cellular activities. It is part of the broader semaphorin family which has a role in directing axonal growth during development. This protein contributes to the regulation of immune cell activity including the activation and differentiation of T cells. The presence of SEMA4A influences immune responses highlighting its importance in maintaining immune system balance.
Pathways
SEMA4A engages in the immune system and axon guidance pathways. In the immune system it interacts with proteins like Plexin B1 and CD72 influencing T cell proliferation and function. Through the axon guidance pathway SEMA4A helps direct the growth of nerve fibers impacting neuronal circuit formation. These interactions exemplify SEMA4A's role in linking cell signaling with physiological outcomes.
