SEMA4F also known as Semaphorin-4F is a member of the semaphorin family of proteins. It has an approximate molecular mass of 85 kDa. This protein is expressed mainly in the central nervous system particularly in neurons and glial cells. SEMA4F is a membrane-bound protein that plays a role in axonal guidance interacting with specific cell surface receptors during neural development.
SEMA4F serves as a mediator in cell signaling processes critical for neuronal connections. It is part of a larger signaling complex that includes plexins and neuropilins which help guide axonal pathfinding and neuronal wiring. SEMA4F influences neurite outgrowth and contributes to the establishment of neural networks by regulating cytoskeletal dynamics and cell adhesion properties.
Several cellular signaling mechanisms involve SEMA4F interactions. SEMA4F is involved in the PI3K/Akt and MAPK/ERK pathways both essential for cell growth and survival. Within these pathways SEMA4F interacts with Plexin-B1 which transduces intracellular signals affecting cytoskeleton reorganization. These interactions highlight its role in neurodevelopment and differentiation.
SEMA4F has associations with neurodegenerative diseases and cancer. Aberrant SEMA4F expression has links to Alzheimer's disease where it interacts with proteins like Β-amyloid precursor protein potentially influencing disease progression. In cancer biology abnormal SEMA4F signaling is observed in gliomas with its interactions with Plexin-B1 potentially affecting tumor growth and metastasis.
