Bifunctional axonal guidance cue regulated by sulfated proteoglycans; attractive effects result from interactions with heparan sulfate proteoglycans (HSPGs), while the inhibitory effects depend on interactions with chondroitin sulfate proteoglycans (CSPGs) (By similarity). SEMA5A also known as Semaphorin-5A is a member of the semaphorin family of proteins which guide axonal development and cell migration. This protein has a mass of approximately 89 kDa. SEMA5A is expressed in various tissues including the brain where it plays a significant part in neuronal development. It is a transmembrane protein with specific structural domains that facilitate its role in cell signaling affecting axonal guidance and cellular movement.
SEMA5A functions in cell signaling mechanisms essential for proper neural circuit formation and development. The protein interacts with receptors on the cell surface including Plexin-B3 which serves as one of its main receptors and this interaction mediates axonal guidance. Not typically part of a larger complex SEMA5A still influences multiple biological processes such as cell adhesion and migration affecting how neurons connect and communicate in the central nervous system.
SEMA5A operates within important signaling pathways like the semaphorin signaling pathway and the axon guidance pathway. These pathways play significant roles in neural development and involve key interactions with proteins like Plexins and Neuropilins. Within these pathways SEMA5A contributes to the establishment of neural connections and synaptic stability interacting with related proteins that direct cellular growth and structure formation.
Research associates SEMA5A with conditions such as autism spectrum disorder and certain types of cancer. Alterations in SEMA5A expression or function can contribute to neural connectivity irregularities observed in autism. Additionally SEMA5A shows involvement in cancer cell migration and invasion.
