SEMA5A also known as Semaphorin-5A is a member of the semaphorin family of proteins which guide axonal development and cell migration. This protein has a mass of approximately 89 kDa. SEMA5A is expressed in various tissues including the brain where it plays a significant part in neuronal development. It is a transmembrane protein with specific structural domains that facilitate its role in cell signaling affecting axonal guidance and cellular movement.

Bifunctional axonal guidance cue regulated by sulfated proteoglycans; attractive effects result from interactions with heparan sulfate proteoglycans (HSPGs), while the inhibitory effects depend on interactions with chondroitin sulfate proteoglycans (CSPGs) (By similarity). In glioma cells, SEMA5A stimulation of PLXNB3 results in the disassembly of F-actin stress fibers, disruption of focal adhesions and cellular collapse as well as inhibition of cell migration and invasion through ARHGDIA-mediated inactivation of RAC1.
