SEPP1 was immunoprecipitated from 0.35 mg HepG2 (human hepatocellular carcinoma epithelial cell), whole cell lysate with ab277526 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab277526 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/5000 dilution.

SEPP1 also known as Selenoprotein P is a glycoprotein characterized by multiple selenocysteine residues which play a critical role in its function. SEPP1 has a molecular mass of approximately 60 kDa. It expresses predominantly in the liver but can be found in other tissues like the brain kidney and plasma. In the liver it is involved in selenium transport and homeostasis carrying selenium to other tissues through the blood.
Biological function summary

SEPP1 acts as an antioxidant helping to defend cells against damage from reactive oxygen species. It exists as part of a complex system within the body to balance oxidative stress. Moreover SEPP1 contributes to the recycling of selenium in the body ensuring adequate supply for selenoprotein synthesis in various tissues. It serves essential functions in neurological health and immune response leveraging its selenium content to protect cells and maintain cellular activities.

Pathways

SEPP1 is significantly involved in selenium homeostasis and the oxidative stress response pathways. It interacts with proteins like glutathione peroxidase (GPx) and thioredoxin reductase which also depend on selenium for their antioxidant activities. The involvement of SEPP1 in these pathways is critical for mitigating oxidative damage and supporting the function of other selenium-dependent enzymes essential for maintaining cellular health.

SEPP1 is associated with neurodegenerative diseases and certain types of cancer.
