Function

Filament-forming cytoskeletal GTPase. Required for normal organization of the actin cytoskeleton. Involved in cytokinesis. May play a role in HCV RNA replication. Forms a filamentous structure with SEPTIN12, SEPTIN6, SEPTIN2 and probably SEPTIN4 at the sperm annulus which is required for the structural integrity and motility of the sperm tail during postmeiotic differentiation (PubMed:25588830).

Activity summary

SEPT6 also known as Septin-6 is a member of the septin family of GTP-binding proteins with a molecular mass of approximately 48 kDa. It forms part of the cytoskeleton and plays a role in cellular functions like cytokinesis and cell cycle regulation. SEPT6 is expressed widely in human tissues including the brain and testis. The protein interacts with other septin family members contributing to its structural role in cells.

Biological function summary

The septin family including SEPT6 is important for cellular processes such as exocytosis and vesicle trafficking. SEPT6 forms complexes with other proteins joining septins like SEPT2 and SEPT7 contributing to the maintenance of cellular architecture. Its interaction with cell membranes allows for the formation of diffusion barriers necessary in various cellular regions and phases.

Pathways

The septin network with SEPT6 plays significant roles in signal transduction and the regulation of cell division. The protein is integral to the MAPK/ERK pathway which controls cellular activities like growth. It closely relates to the interactions with CDC42 a small GTPase essential for actin cytoskeleton organization. SEPT6's contribution to phagocytosis and polarization exemplifies the importance of septins in cellular dynamics.

Associated diseases and disorders

SEPT6 often associates with conditions like cancer and neurodegenerative diseases. Its dysregulation or mutation may lead to abnormal cell growth contributing to tumorigenesis. In the brain abnormal expression of SEPT6 connects with proteins like tau in neurodegenerative diseases indicating a role for SEPT6 in pathologies involving impaired cellular structures. Proper functioning of SEPT6 therefore maintains cellular integrity and its disruption may be a biomarker or therapeutic target in certain disease contexts.
