Activity summary

Serotonin also known as 5-hydroxytryptamine (5-HT) is a neurotransmitter mainly involved in modulating mood emotion and behavior. It weighs approximately 176.2 Da and appears in several body tissues including the central nervous system gastrointestinal tract and blood platelets. Neurons within the raphe nuclei of the brainstem predominantly synthesize serotonin influencing various physiological processes. Its mechanical role involves binding to and activating specific serotonin receptors classified into seven different types each mediating distinct signaling pathways.

Biological function summary

Serotonin regulates multiple bodily functions beyond mood such as appetite sleep and circadian rhythm. It forms part of a complex system interacting with various receptor subtypes which include the anti-serotonergic pathways inhibiting its action. Each receptor subtype leads to different downstream effects in cells contributing to the diversity of serotonin's biological roles.     1 ug/mL Serotonin-crosslink-BSA was coated onto 96-wells. Serial dilution of free Serotonin (0, 19.53125, 39.0625, 78.125, 156.25, 312.5, 625, 1250, 2500, 5000 ng/mL) was added (50 uL). This was followed by adding 50 ng/mL biotin-labeled ab317525 (50 uL) into each well. The plate was incubated for 30 mins. After washing, 1:20000 diluted SA-HRP was added and incubated for 30 mins. After washing, 100 uL of TMB was added and incubated for 5-10 mins without shaking for color development. Then 100 uL of stop solution (4.9% Orthophosphoric acid) was dispensed and OD was read at 450nm on a microplate reader within 5-10 mins.
