SerpinB3 also known as Squamous Cell Carcinoma Antigen (SCCA) is part of the serine protease inhibitor family. This protein has a molecular mass of approximately 45 kDa. It is expressed in various human tissues including skin liver and the respiratory epithelium. SerpinB3 acts by inhibiting cysteine proteinases through binding to them which helps regulate their activity and prevent excessive proteolysis. Such regulatory mechanisms are essential for maintaining cellular homeostasis.
SerpinB3 plays a role in cellular proliferation differentiation and protection against apoptosis. It is not reported as part of a specific protein complex but it engages in interactions that influence cell signaling pathways. By protecting cells from programmed cell death SerpinB3 can contribute to tissue repair and regeneration. Its expression levels can change significantly during cell stress responses suggesting its involvement in adaptive processes to environmental stimuli.
Protein SerpinB3 is involved in inflammatory and immune pathways. It influences the NF-κB signaling pathway which is important for immune response regulation and inflammation. This pathway involves multiple proteins including IκB which acts as an inhibitor and other cytokines and transcription factors that facilitate signal transduction in response to stress. SerpinB3’s modulation of these pathways reflects how it affects both innate and adaptive immune responses.
SerpinB3 has been associated with the progression of squamous cell carcinoma and liver fibrosis. In squamous cell carcinoma its interaction with proteases like cathepsin contributes to cancer cell survival and metastasis. For liver fibrosis SerpinB3’s inhibition of apoptosis in hepatocytes may lead to excessive tissue scarring. These connections illustrate its potential as a biomarker for diagnosis and as a target for therapeutic intervention.
