SESN3 also known as Sestrin 3 is a protein involved in cellular stress response with a mass of approximately 57 kDa. This protein works as an antioxidant and helps in regulating the cellular redox state. SESN3 is expressed in various tissues most prominently in metabolically active tissues such as the liver and muscle. It responds to oxidative stress by influencing antioxidant defense mechanisms and also maintains cellular homeostasis.
SESN3 plays an important role in metabolic regulation and cellular protection. It activates AMP-activated protein kinase (AMPK) contributing to the suppression of the mammalian target of rapamycin complex 1 (mTORC1) signaling. SESN3 does not exist as a part of a larger protein complex itself but interacts with other proteins to exert its functions. Through these interactions SESN3 helps manage cellular metabolism preventing damage from oxidative stress.
SESN3 participates in the PI3K/AKT/mTOR and AMPK signaling pathways both essential for regulating cell growth and energy metabolism. It interacts with proteins such as mTOR and TSC2 (tuberous sclerosis complex 2) which are necessary for balancing cellular growth and nutrient availability. SESN3 modulates these pathways by inhibiting mTORC1 activity impacting processes like protein synthesis and autophagy.
SESN3 has associations with metabolic disorders and cancer. Dysregulation of SESN3 affects pathways like AMPK and mTOR linked to type 2 diabetes and obesity. Additionally changes in SESN3 levels influence tumor growth as SESN3 affects processes involved in cell survival and proliferation. SESN3's interaction with AMPK and mTOR links it to cancerous processes highlighting its potential as a therapeutic target.
