Component of the 17S U2 SnRNP complex of the spliceosome, a large ribonucleoprotein complex that removes introns from transcribed pre-mRNAs (PubMed:10882114, PubMed:11533230, PubMed:32494006, PubMed:34822310, PubMed:8022796). The 17S U2 SnRNP complex (1) directly participates in early spliceosome assembly and (2) mediates recognition of the intron branch site during pre-mRNA splicing by promoting the selection of the pre-mRNA branch-site adenosine, the nucleophile for the first step of splicing (PubMed:10882114, PubMed:11533230, PubMed:32494006, PubMed:34822310). Within the 17S U2 SnRNP complex, SF3A3 is part of the SF3A subcomplex that contributes to the assembly of the 17S U2 snRNP, and the subsequent assembly of the pre-spliceosome 'E' complex and the pre-catalytic spliceosome 'A' complex (PubMed:10882114, PubMed:11533230). Involved in pre-mRNA splicing as a component of pre-catalytic spliceosome 'B' complexes (PubMed:29360106, PubMed:30315277).

SF3A3 also known as Splicing Factor 3A Subunit 3 plays an important role in pre-mRNA splicing. It has a mass of approximately 60 kDa. SF3A3 is a component of the splicing machinery and localizes predominantly to the nucleus where it is involved in the assembly of the spliceosome. The gene encoding SF3A3 is expressed in a wide range of tissues indicating its fundamental role in the splicing process across different cell types.
SF3A3 is part of the U2 small nuclear ribonucleoprotein (snRNP) complex which is important for the second step of the splicing process. This complex recognizes the pre-mRNA branch site and participates in the assembly of the spliceosome a complex that excises introns and joins exons to produce mature mRNA. The activity of SF3A3 is essential for the accurate splicing of precursor mRNA transcripts impacting gene expression regulation.
SF3A3 plays a role in the mRNA processing pathway and the spliceosome pathway. These pathways are vital for gene expression regulation and the generation of functional RNA products. Within the spliceosome pathway SF3A3 is associated with other splicing factors such as SF3B1 which also plays a role in splicing regulation and influences the assembly and function of the spliceosome complex.
SF3A3 has been linked to cancer and myelodysplastic syndromes. Mutations or dysregulation of SF3A3 and its associated splicing factors like SF3B1 can lead to aberrant splicing which may contribute to oncogenesis and disease progression. Understanding SF3A3's role in these disorders could provide insights into therapeutic targets for treatment strategies.
