Supplementary information

SGT1/ECD also known as SGT1 (Suppressor of G2 allele of SKP1) or ECD (Ectodermal Dysplasia protein) is a regulatory protein with a mass of approximately 36 kDa. It is highly expressed in various eukaryotic cells and tissues. This protein plays an important mechanical role by facilitating the assembly and stabilization of protein complexes. SGT1/ECD acts as a co-chaperone assisting HSP90 in its function which is critical for client protein folding and activity.
Biological function summary

SGT1/ECD participates in the assembly of multi-protein complexes. It is also intertwined with the SCF (SKP1-Cullin1-F-box) ubiquitin ligase complex where it contributes to substrate recognition and proteasomal degradation. Furthermore SGT1/ECD is important in regulating the cell cycle and signal transduction processes involving plant immunity and kinetochore function during chromosome segregation.

Pathways

SGT1/ECD engages in both the plant and animal kingdoms. In plants it plays a role in the resistance (R) gene-mediated signaling pathways and the jasmonic acid pathway where it pairs with chaperones like HSP90. In animals it links with cell cycle pathways affecting proteins such as SKP1 and HSP90. Through these interconnected pathways SGT1/ECD is linked to vital signaling mechanisms impacting cellular responses and stress tolerance.

SGT1/ECD shows connections to cancer and immune response dysregulation. Within cancer biology altered SGT1/ECD expression influences the stability of oncogenic client proteins via HSP90. Additionally in the context of immune disorders it is involved in plant immunity defects where malfunction in SGT1/ECD-mediated pathways can lead to impaired disease resistance. Its associations with proteins like HSP90 and components of the SCF complex highlight its relevance in maintaining cellular homeostasis and proper immune functions.

Target data

Regulator of p53/TP53 stability and function. Inhibits MDM2-mediated degradation of p53/TP53 possibly by cooperating in part with TXNIP (PubMed : 16849563, PubMed : 23880345). May be involved transcriptional regulation. In vitro has intrinsic transactivation activity enhanced by EP300. May be a transcriptional activator required for the expression of glycolytic genes (PubMed : 19919181, PubMed : 9928932). Involved in regulation of cell cycle progression. Proposed to disrupt Rb-E2F binding leading to transcriptional activation of E2F proteins (PubMed : 19640839). The cell cycle -regulating function may depend on its RUVBL1-mediated association with the R2TP complex (PubMed : 26711270). May play a role in regulation of pre-mRNA splicing (PubMed : 24722212). Participates together with DDX39A in mRNA nuclear export (PubMed : 33941617).
