SH2D1A also known as SLAM-associated protein (SAP) is a small intracellular protein with a molecular weight of approximately 15 kDa. It is primarily expressed in T cells and natural killer (NK) cells playing important role in immune signaling. SAP contains SH2 domains that facilitate its interaction with signaling lymphocytic activation molecule (SLAM) family receptors. These interactions mediate downstream signaling cascades that control immune responses.
Biological function summary

The SAP protein influences T cell and NK cell activity by modifying signaling pathways involved in cell development and function. SAP does not function alone; it is part of a signaling complex with SLAM and other adaptor molecules. This complex regulates cytokine production and cytotoxic activity which are important for maintaining effective immune responses against infections.

Pathways

SH2D1A is an important component in the SLAM signaling pathway which interacts closely with the ITK signaling pathway. These pathways are essential for proper development and function of T cells and NK cells. SAP also modulates interactions with proteins such as Fyn a tyrosine kinase linked to T cell receptor signaling. Through these pathways it influences cellular processes that ensure effective immune surveillance and response.

SH2D1A mutations or dysfunction are directly connected to X-linked lymphoproliferative disease (XLP1) a condition characterized by the inability to regulate immune responses often resulting in severe and life-threatening infections. The protein is associated with immune dysregulation observed in the disease contributing to defective signaling in T cells and NK cells. Other disorders related to SH2D1A dysfunction include certain lymphomas where aberrant expression of this and related proteins such as SLAM receptors affect cellular proliferation and immune surveillance.

Target data

Cytoplasmic adapter regulating receptors of the signaling lymphocytic activation molecule (SLAM) family such as SLAMF1, CD244, LY9, CD84, SLAMF6 and SLAMF7. In SLAM signaling seems to cooperate with SH2D1B/EAT-2. Initially it has been proposed that association with SLAMF1 prevents SLAMF1 binding to inhibitory effectors including INPP5D/SHIP1 and PTPN11/SHP-2 (PubMed : 11806999). However, by simultaneous interactions, recruits FYN which subsequently phosphorylates and activates SLAMF1 (PubMed : 12458214). Positively regulates CD244/2B4- and CD84-mediated natural killer (NK) cell functions. Can also promote CD48-, SLAMF6 -, LY9-, and SLAMF7-mediated NK cell activation. In the context of NK cell-mediated cytotoxicity enhances conjugate formation with target cells (By similarity). May also regulate the activity of the neurotrophin receptors NTRK1, NTRK2 and NTRK3.
