The SH3 domain-containing kinase-binding protein 1 (SH3KBP1) also known as Cin85 is a multifunctional adapter protein involved in signal transduction pathways. It has a molecular mass of approximately 85 kDa. SH3KBP1 is expressed in a variety of tissues including the brain heart and lungs reflecting its diverse roles in cellular processes. This protein contains multiple SH3 domains that facilitate the binding with other proteins making SH3KBP1 an important modulator of protein-protein interactions.
Biological function summary

SH3KBP1 plays a role in cellular signaling and endocytosis. It forms part of a complex that participates in the regulation of membrane trafficking where it helps in internalization and down-regulation of receptor tyrosine kinases. SH3KBP1 interacts with other signaling molecules allowing it to coordinate the spatial and temporal dynamics of signaling pathways. This protein’s involvement in endocytic pathways highlights its significance in regulating cellular responses to external stimuli.

Pathways

SH3KBP1 is an integral part of the epidermal growth factor receptor (EGFR) signaling and the B-cell receptor (BCR) signaling pathways. Adapter protein involved in regulating diverse signal transduction pathways. Involved in the regulation of endocytosis and lysosomal degradation of ligand-induced receptor tyrosine kinases, including EGFR and MET/hepatocyte growth factor receptor, through an association with CBL and endophilins. The association with CBL, and thus the receptor internalization, may be inhibited by an interaction with PDCD6IP and/or SPRY2. Involved in regulation of ligand-dependent endocytosis of the IgE receptor. Attenuates phosphatidylinositol 3-kinase activity by interaction with its regulatory subunit (By similarity). May be involved in regulation of cell adhesion; promotes the interaction between TTK2B and PDCD6IP. May be involved in the regulation of cellular stress response via the MAPK pathways through its interaction with MAP3K4. Is involved in modulation of tumor necrosis factor mediated apoptosis.
