Supplementary information

SH3YL1 also known as SH3 domain containing Ysc84-like protein 1 is a protein with a molecular mass of around 42 kDa. Mechanically SH3YL1 acts as a cytoplasmic adapter protein engaging in protein-protein interactions through its SH3 domain which is a common protein interaction domain found in many proteins involved in signal transduction. SH3YL1 expression occurs mainly in neural tissues but it can be found in other tissues at varying levels.
Biological function summary

SH3YL1 participates in intracellular signaling processes that are essential for cell shape and movement. While not part of a large multi-protein complex it interacts with proteins involved in actin cytoskeletal dynamics. Its role in modulating the actin cytoskeleton influences processes like cell adhesion and migration making it an important player in developmental biology and tissue organization.

Pathways

SH3YL1 integrates into signaling pathways that regulate actin cytoskeleton rearrangements such as the Rac1-WAVE2 signaling pathway. In this pathway SH3YL1 interacts with WAVE2 a member of the WAVE regulatory complex to promote actin polymerization. These interactions highlight its relevance in cellular processes such as endocytosis and membrane trafficking linking it to other signaling molecules like Rac1 which further modulates the actin cytoskeleton through related pathways.

SH3YL1 exhibits connections to disorders involving aberrant cell migration and invasion such as cancer metastasis. Increased SH3YL1 activity correlates with enhanced invasive potential in certain cancer cells which may involve its interaction with proteins like WAVE2 in altering cytoskeletal dynamics. Further studies could show a clearer role in neurodegenerative diseases where abnormal cytoskeleton regulation plays a critical role in disease progression.
